The invention provides methods for the enhanced intracellular delivery of therapeutically active molecules to a target tumor cell. In a preferred embodiment, the method comprises providing engineered cells that express a heterologous nucleic acid that encodes a connexin and that contain a pro-drug activating gene that, when expressed, converts a nontoxic substrate to a therapeutic toxic metabolite, contacting target tumor cells in a solid tumor with engineered cells that form functional gap junctions with said target tumor cells, and exposing the engineered and tumor cells to the nontoxic substrate, whereby the nontoxic substrate is converted to a therapeutic molecule such that the therapeutic molecule passes through a gap junction to a target tumor cell.

Procédés permettant d'améliorer l'apport intracellulaire de molécules thérapeutiquement actives à une cellule tumorale cible. Dans un mode de réalisation préféré, ledit procédé consiste à obtenir des cellules recombinées qui expriment un acide nucléique hétérologue codant une connexine et qui contiennent un gène activant un pro-médicament qui, lorsqu'il est exprimé, convertit un substrat non toxique en un métabolite toxique thérapeutique, à mettre en contact des cellules tumorales cibles présentes dans une tumeur solide avec des cellules recombinées qui forment des jonctions lacunaires fonctionnelles avec lesdites cellules tumorales cibles, et à exposer lesdites cellules recombinées et tumorales au substrat non toxique, ledit substrat non toxique étant converti en une molécule thérapeutique de manière à ce que cette dernière traverse la jonction lacunaire et pénètre dans une cellule tumorale cible.
