The present invention provides a method for modifying y-secretase activity in a cell or in an organism by de~stabilizing presenilin through inhibition of Fkbp13. Inhibition of Fkbp13 de-stabilizes presenilin and prevents formation of a functional y-secretase complex thus decreasing y-secretase activity and concomitantly decreasing production of amyloid-~ peptides. The invention also provides methods for identifying agents which function to inhibit Fkbp13.

La présente invention concerne une méthode de modification de l'activité de .gamma.-secrétase dans une cellule ou un organisme par déstabilisation de la préséniline par inhibition de Fkbp13. Cette inhibition de Fkbp13 permet de déstabiliser la préséniline et de prévenir la formation d'un complexe de .gamma.-secrétase fonctionnel, ce qui permet de diminuer l'activité de .gamma.-secrétase et, de manière concomitante, la production de peptides .beta. amyloïdes. Ladite invention a aussi pour objet des méthodes d'identification d'agents qui agissent afin d'inhiber Fkbp13.
