A method of modifying a nucleotide chain comprising treating the nucleotide chain (I) to be modified, which has a nucleotide sequence carrying a specific base such as hypoxanthine (Hx) in the 3~-side, with an enzyme specifically digesting the nucleotide having the specific base as described above, imparting a functional group (for example, an aldehyde group) capable of binding to a desired modification substance (for example, NH2R having an amino group) to the 3~-end of the nucleotide chain (I), and then attaching the modification substance to the 3~-end of the nucleotide chain. In this method, the nucleotide chain having the nucleotide sequence carrying a specific base that serves as the substrate of the enzyme at the 3~-side is employed for the subject to be modified. Thus, the nucleotide sequence moiety alone is digested to thereby form a functional group that reacts with a desired modification substance and binds thereto. Owing to this process, the nucleotide chain can be directly modified with the modification substance and, therefore, can be easily labeled or tagged. In the case where the nucleotide chain is to be immobilized, moreover, the modification substance can be stabilized as a linker and thus rigid immobilization can be established.
L'invention concerne un procédé de modification d'une chaîne nucléotide consistant à traiter la chaîne nucléotide à modifier (I), présentant une séquence porteuse d'une base spécifique, telle que l'hypoxanthine (Hx) en position latérale 3', avec une enzyme digérant spécifiquement le nucléotide porteur de la base spécifique précitée, à impartir un groupe fonctionnel (par exemple, un groupe aldéhyde) capable de liaison à une substance de modification souhaitée (par exemple, NH¿2?R ayant un groupe amino) à l'extrémité 3' de la chaîne nucléotide (I), puis à lier la substance de modification à l'extrémité 3' de la chaîne nucléotide. Dans ce procédé, la chaîne nucléotide porteuse d'une base spécifique servant de substrat de l'enzyme en position latérale 3' est employée pour le sujet à modifier. Ainsi, seule la fraction séquence nucléotide est digérée de manière à former un groupe fonctionnel réagissant avec une substance de modification désirée et se lie à celle-ci. Grâce à ce procédé, la chaîne nucléotide peut être directement modifiée avec la substance de modification et, de ce fait, peut être facilement étiquetée ou marquée. En outre, dans le cas où la chaîne nucléotide doit être immobilisée, la substance de modification peut être stabilisée sous forme d'un lieur, ce qui permet d'établir une immobilisation rigide.
