The present invention relates to a process of preparing benzisoxazole methane sulfonic acid-chloride (BOS-CI) as an zonisamide intermediate via chlorination of benzisoxazole methane sulfonate. The present invention also disloses a process of preparing zonisamide via amidation of BOS-CI. More particularly, the present invention provides a process of preparing zonisamide, comprising the steps of : a) chlorinating BOS; salts or esters thereof, with SOCl2 in an organic solvent and/or in the presence of a catalyst to form BOS-Cl; and b) amidating BOS-Cl in the presence of ammonia selected from the group consisting of aqueous ammonia in a biphasic system, masked ammonia and dry ammonia to form zonisamide.
La présente invention concerne un procédé de préparation de chlorure d'acide benzisoxazole méthane sulfonique (BOS-Cl) comme intermédiaire zonisamide, par chloration de benzisoxazole méthane sulfonate. La présente invention concerne également un procédé de préparation d'un zonisamide par amidation du BOS-Cl. L'invention concerne, plus précisément, un procédé de préparation d'un zonisamide, consistant : a) à chlorer le BOS, ses sels ou ses esters, avec du SOCl¿2? dans un solvant organique et/ou en présence d'un catalyseur pour former du BOS-Cl ; et b) à réaliser l'amidation du BOS-CL en présence d'ammoniac sélectionné dans le groupe comprenant de l'ammoniac aqueux dans un système biphasique, de l'ammoniac masqué et de l'ammoniac anhydre, pour former un zonisamide.
