The invention relates to a method for preparing nucleotides or monomer nucleotide analogues, which comprises the following steps: (1) bonding a soluble polyethyleneglycol substrate including at least one diacid or ether-acid bonding arm and a nucleoside or monomer nucleoside analogue to an amino or hydroxyl group of the nucleoside using a bonding agent; (2) at least one phosphorylation step of said nucleoside or nucleoside analogue bonded to said substrate using a phosphorylation agent; (3) cleaving said substrate and recovering at least one nucleotide or monomer nucleotide analogue. The invention also relates to said nucleotides.
L'invention concerne un procédé de préparation de nucléotides ou analogue de nucléotides monomères comportant les étapes de : (1) couplage d'un support polyéthylèneglycol soluble pourvu d'au moins un bras de liaison diacide ou éther-acide et d'un nucléoside ou analogue de nucléoside monomère, sur un groupe aminé ou hydroxyle du nucléoside à l'aide d'un agent de couplage; (2) au moins une étape de phosphorylation dudit nucléoside ou analogue de nucléoside couplé audit support avec un agent de phosphorylation; (3) clivage dudit support et récupération d'au moins un nucléotide ou analogue de nucléotide monomère. Les nucléotides préparés sont des outils biologiques.
