There is provided a method for growing human intervertebral cells. Disc tissue is surgically removed from a normal disc of a patient, the cells expanded by feeding with a cell stimulant such as a growth factor, or a cytokine or a bioactive agent to form monolayer primary cell cultures on a plastic mesh such as a nylon mesh. In the case of a growth factor, fetal bovine serum is preferred as it improves cell proliferation and production of appropriate extracellular matrix components. In another aspect of this invention, the monolayer primary cell cultures are seeded in alginate or agarose and fed again with the cell stimulant until three-dimensional cell cultures are formed. The cells are recovered from the alginate or agarose or from monolayer cultures. Re-implantation is carried out using bioresorbable carriers or cell suspensions.

L'invention concerne une méthode de culture de cellules intervertébrales humaines. On effectue une intervention chirurgicale sur un patient pour extraire du tissu discal d'un de ses disques intervertébraux normaux. On développe ces cellules en les nourrissant avec un stimulant cellulaire tel qu'un facteur de croissance, une cytokine ou un agent bioactif, de façon à former des cultures monocouche de cellules primaires sur une toile plastique, telle qu'une toile de nylon. Lorsqu'on utilise un facteur de croissance, on préfère le sérum de foetus bovin, étant donné qu'il améliore la prolifération cellulaire et la production de composants matriciels extracellulaires appropriés. Selon un autre aspect de cette invention, les cultures monocouche de cellules primaires sont ensemencées dans un alginate ou un agarose, puis nourris à nouveau avec le stimulant cellulaire jusqu'à formation de cultures cellulaires tridimensionnelles. On récupère les cellules de l'alginate ou de l'agarose, ou encore des cultures monocouche. On réalise la réimplantation à l'aide de vecteurs ou de cellules en suspension biorésorbables.
