The invention relates to a method for the chemical synthesis of interferon proteins containing one or more cysteine residues, by the native chemical ligation of segments of the full length interferon protein. The segments of the protein are selected based on the position of the cysteine residues in the interferon protein, with the position of the cysteine residue as a potential site of native chemical ligation. The full-length, fully deprotected polypeptide is folded into biologically active interferon protein by oxidation of the cysteine residues. The invention further relates to the production of the individual peptide fragments which act as intermediates in the synthesis of interferon-alpha. The present invention still further relates to groups of such peptide intermediates which can be utilized together to produce interferon-alpha and interferon-alpha-like proteins.
L'invention se rapporte à un procédé pour la synthèse chimique de protéines interféron contenant au moins un résidu de cystéine, par ligature chimique native de segments de la protéine interféron pleine longueur. Ces segments de la protéine sont sélectionnés selon la position des résidus de cystéine dans la protéine interféron, la position des résidus de la cystéine servant de site potentiel de ligature chimique native. Le polypeptide pleine longueur entièrement déprotégé est plié en une protéine interféron biologiquement active par oxydation des résidus de cystéine. L'invention se rapporte en outre à la production de fragments peptidiques individuels qui agissent en tant qu'intermédiaires dans la synthèse d'interféron alpha. L'invention se rapporte en outre à des groupes de ces intermédiaires peptidiques qui peuvent être utilisés ensemble pour produire des interférons alpha et des protéines similaires.
