A method for genetically engineering cells to produce soluble and secretable Golgi processing enzymes instead of naturally occurring membrane-bound enzymes. Cells are genetically engineered to express glycosyltransferases which lack both a membrane anchor and a retention signal. The resulting altered enzyme becomes soluble and secretable by the cell without losing its catalytic activity. Secretion of the soluble glycosyltransferase by the cell provides for increased production and simplified recovery of glycosyltransferase.
Procédé de manipulation génétique de cellules permettant de produire des enzymes de traitement de Golgi solubles et sécrétables remplaçant les enzymes naturelles liées à la membrane. Des cellules sont manipulées génétiquement pour exprimer des glycosyltransférases manquant à la fois d'un ancrage de membrane et d'un signal de rétention. L'enzyme modifiée obtenue devient soluble et sécrétable par la cellule sans perdre son activité catalytique. La sécrétion de glycosyltransférase soluble par la cellule permet une plus grande production et une récupération simplifiée de glycosyltransférase.
