The present invention relates to a method for the isolation of proteins that comprise disulfide-bonds in their native conformation. Essentially, a method of the present makes the use of reducing agents such as .beta.-mercaptoethanol or dithiothreitol in protein isolation methods obsolete. A method of the present invention is particularly suitable for the isolation of precursor proteins such as proinsulin from recombinant cells.
L'invention concerne une méthode d'isolation de protéines qui comprennent des liaisons disulfures dans leur conformation native. De manière générale, la méthode de l'invention rend obsolète la mise en oeuvre de procédés faisant intervenir des agents de réduction de type .beta.-mercaptoéthanol ou dithiothréitol dans l'isolation de protéines. Ladite méthode s'avère particulièrement appropriée à l'isolation de protéines précurseurs de type proinsulines à partir de cellules de recombinaison.
