The present disclosure relates to method of DNA amplication with a first primer that has a random sequence of nucleotides as its 3' end and a generic sequence 5' of the random nucleotides, as well as a second primer with the generic sequence of the first primer. The disclosure further relates to a method relates to a method of participating DNA on a solid medium to improve DNA amplification. In a preferred embodiment, the presently disclosed methods are used for high-throughput genotyping of DNA samples, such as bloodstains or trace amounts of DNA.

L'invention concerne des procédés d'amplification de l'ADN mettant en oeuvre une première amorce possédant une séquence aléatoire de nucléotides au niveau de son extrémité 3' et une séquence générique 5' des nucléotides aléatoires, ainsi qu'une seconde amorce comprenant la séquence générique de la première amorce. L'invention concerne également un procédé consistant à précipiter un ADN sur un support solide de manière à améliorer l'amplification de l'ADN. Dans un mode de réalisation préféré, les procédés sont mis en oeuvre aux fins d'obtention d'un génotypage à rendement élevé d'échantillons d'ADN, tels que des tâches sang ou des quantités à l'état de trace d'ADN.
