The invention concerns a method for transferring into a mammal's oocyte a site-specific, in particular Cre-mediated, recombinase in its active form. Said recombinase is expressed under the control of a specific promoter in male germinal cells comprising at least a specific site and remains associated with the spermatozoid chromatin. Said method is particularly useful for remodelling an animal genome, for instance for inserting a foreign sequence into an animal genome, and for replacing a sequence by another. In the event where said promoter is a Sycp 1 promoter, an application mode enables to recombine paternal and maternal chromosomes. The invention also concerns animals obtainable by said method, and the use of said transgenic animals in pharmaceutical, cosmetic or agri-food industries.
La présente invention concerne un procédé pour transférer dans un oocyte d'un mammifère une recombinase spécifique de site, en particulier Cre, sous sa forme active. Ladite recombinase est exprimée sous le contrôle d'un promoteur spécifique dans des cellules germinales mâles comportant au moins un site spécifique et reste associée à la chromatine des spermatozoïdes. Ce procédé est particulièrement utile pour remodeler le génome d'un animal, par exemple pour insérer une séquence étrangère dans le génome d'un animal, et pour remplacer une séquence par une autre. Dans le cas où ledit promoteur est le promoteur Sycp1, un mode d'application permet de réaliser des recombinaisons entre chromosomes paternels et maternels. La présente invention porte également sur les animaux susceptibles d'être obtenus par ce procédé, et sur l'utilisation de ces animaux transgéniques dans les industries pharmaceutiques, cosmétiques, et agro-alimentaire.
