The invention is a method for reproducing Douglas-fir by somatic embryogenesis using plant tissue culture techniques in a multistage culturing process. A suitable explant, typically the fertilized embryo excised from an immature seed, is first cultured on a medium that induces multiple early stage embryos. These are multiplied in a second culture having reduced growth hormones where the early stage embryos grow in size and vigor to advanced early stage embryos. The embryos may then be transferred direclty to a cotyledonary embryo development culture containing an adsorbent such as activated charcoal. Preferably, they are first singulated by shaking in two or more liquid subcultures containing the adsorbent prior to placing them on the development medium. After several weeks somatic embryos having the appearance of zygotic embryos will have formed. These may be germinated before or after storage and transplanted to soil for further growth.
L'invention concerne un procédé qui permet de reproduire le Douglas taxifolié par embryogenèse somatique à l'aide de techniques de culture de tissus végétaux dans le cadre d'un processus de culture à plusieurs étapes. On cultive d'abord un explant approprié, en général l'embryon fécondé excisé à partir d'une graine immature, dans un milieu qui induit plusieurs embryons en phase précoce qu'on multiplie dans une deuxième culture à teneur en hormones de croissance réduite où ils se développent en taille et en vigueur pour devenir des embryons avancés de phase précoce. On peut alors les transférer directement vers une culture de développement d'embryons cotylédonés qui contient un adsorbant tel que du charbon actif. On les divise d'abord de préférence, en les agitant dans deux sous-cultures liquides ou plus qui contiennent cet adsorbant, avant de les placer dans le milieu de développement. Des embryons somatiques ayant l'apparence d'embryons zygotiques se forment après quelques semaines et on peut les faire germer avant ou après entreposage puis les transplanter dans le sol où ils grandiront.
