This invention comprises engineered cells that express a nucleic acid that encodes a calbindin molecule, and exhibit the ability to secrete insulin in a glucose-sensitive fashion. This invention also comprises an artificial tissue that, when implanted into recipients that do not secrete insulin in a glucose- sensitive fashion, imparts to the recipients the ability to secrete insulin in a glucose-sensitive fashion. The artificial tissue comprises engineered cells that express a nucleic acid that encodes a calbindin molecule and secrete insulin in a glucose-sensitive fashion, enclosed by a semipermeable porous matrix.
L'invention porte sur des cellules transformées génétiquement exprimant un acide nucléique codant pour une molécule de calbindine et susceptible de sécréter de l'insuline en réponse à la présence de glucose. L'invention porte également sur un tissu artificiel qui implanté dans un patient ne sécrétant pas d'insuline en fonction du glucose présent lui confère la faculté d'en sécréter. Ledit tissu artificiel comporte des cellules transformées génétiquement exprimant un acide nucléique codant pour une molécule de calbindine et sécrétant de l'insuline en fonction du glucose présent. Lesdites cellules sont enrobées d'une matrice poreuse semi-perméable.
