The invention relates to a method for screening modulators of mitochondrial function comprising adding a compound to be tested in a purified, isolated mitochondria preparation and simultaneously using fluorimetric analysis of mitochondrial morphology, and especially real-time fluorimetric analysis, combining analysis of morphometric parameters (SSC/FSC parameters) with analysis of membrane integrity by dye fluorescence. Application to the obtention of peptides which induce mitochondrial membrane permeabilization.
La présente invention concerne une technique de recherche de modulateurs de la fonction mitochondriale qui consiste à ajouter un composé à tester dans une préparation purifiée de mitochondries isolées et à utiliser simultanément une analyse fluorimétrique de morphologie mitochondriale et, plus particulièrement, une analyse fluorimétrique en temps réel combinant l'analyse des paramètres morphométriques (SSC/FSC) avec l'analyse de l'intégrité de membrane par fluorescence. La présente invention concerne une application de l'obtention de peptides qui induit la perméabilisation de membrane mitochondriale.
