ABSTRACT OF THE DISCLOSURE The determination of the activity of an enzyme in a medium is effected by a method in which a substrate which gives a free aromatic primary amine on treatment with the enzyme is contacted with the medium and the amine produced is estimated colorimetrically using as a colour reagent an alkali metal pentacyanoammineferroate which has been treated with an oxidising agent comprising hydrogen perioxide or a salt of an inorganic peracid. The method can be effected using a diagnostic kit containing the substrate, the colour reagent and a buffer.
