A method and device for carrying out immunoassays in which non analyte specific binding of heterophilic antibodies to a labeled antibody in a capture region produces an incorrect measure of the amount of an analyte attached to the antibody. Immunoglobulin from the same animal source as the labeled antibody is added to the sample fluid to prevent non-specific binding of the heterophilic antibodies in the capture region. One part of specific binding pair is added to said antibody or its label capable of binding to a second part of the binding pair immobilized in a control region downstream of said capture region for trapping the portion of the labeled anti-body which is not bound to the analyte. Preferably said binding pair is biotin/avidin or fluoroscein/anti-fluoroscein.
L'invention a trait à un procédé et à un dispositif permettant de réaliser des dosages immunologiques dans lesquels la liaison non spécifique à une substance à analyser d'anticorps hétérophiles à un anticorps marqué dans une zone de capture produit une mesure erronée de la quantité d'une substance à analyser fixée à l'anticorps. On ajoute à l'échantillon de fluide, une immunoglobuline provenant de la même source animale que l'anticorps marqué, afin d'empêcher la liaison non spécifique des anticorps hétérophiles dans la zone de capture. On ajoute audit anticorps ou à son marqueur l'une des parties d'une paire de liaison spécifique, qui est capable de se lier à la seconde partie de la paire de liaison immobilisée dans une zone témoin située en aval de ladite zone de capture, afin de piéger la partie de l'anticorps marqué qui n'est pas liée à la substance à analyser. De préférence, ladite paire de liaison se présente sous la forme biotine/avidine ou fluoroscéine/anti-fluoroscéine.
