The invention relates to the use of arginine as an agent for the extraction of recombinant periplasmic proteins. The invention further relates to a method for the extraction of a periplasmic protein of interest, which consists in: (1) suspending the pellet of cells or cell debris from cells originating from the culture of a prokaryotic microorganism, transformed by means of an expression vector containing a gene coding for the said protein and means for its expression in the periplasm of the said microorganism, in a buffer solution containing arginine; and (ii) recovering the protein of interest in the supernatant of the bacterial suspension thereby obtained.
