This invention is related to a method for the industrial purification of biologically active phycotoxins that comprises providing a suitable amount of a phycotoxin source; if the phycotoxin source is a pellet, said pellet is lysed using suitable methods; the material from the cyanobacterial lysis is subjected to a cold extraction; obtaining a concentrate of the aqueous phase obtained in the previous step or the culture medium of the first step; centrifugating the concentrate to obtain a supernatant; passing the supernatant through a diatomaceous earth column and obtaining a phycotoxin-containing eluate; passing the eluate obtained in the previous step through activated charcoal columns; the eluate from the previous step is passed again through a diatomaceous earth column; the eluate from the previous step is left in an aqueous phase; the partially purified extract from the previous step is subjected to a preparative HPLC in several steps to obtain the pure biologically active phycotoxin.
L'invention concerne un procédé de purification industrielle de phycotoxines biologiquement actives. Ce procédé consiste : à fournir une quantité adéquate d'une source de phycotoxines; si la source de phycotoxines se présente sous forme de pellet, ce dernier est lissé au moyen de procédés appropriés, le matériau issu de la lyse de cyanobactéries est soumis à une extraction à froid; à obtenir un concentré de la phase aqueuse obtenue dans l'étape précédente ou du milieu de culture de la première étape; à centrifuger le concentré afin d'obtenir un surnageant; à faire passer le surnageant dans une colonne de terres à diatomées et à obtenir un élué de la phycotoxine; à faire passer l'élué obtenu dans l'étape précédente dans des colonnes de charbon actif; à faire passer à nouveau l'élué de l'étape précédente dans une colonne de terres à diatomées; à laisser l'élué de l'étape précédente en phase aqueuse; à soumettre l'extrait partiellement purifié de l'étape précédente à une CLHP préparative en plusieurs étapes, ce qui permet d'obtenir la phycotoxine biologiquement active pure.
