The present invention decribes reagents free of detectable contaminating nucleic acids for performing highly sensitive and specific nucleic acids amplification and/or detection. It relates to an improvement in the technology of nucleic acid inactivation prior to nucleic acid testing (NAT) in order to prevent false-positive results. Specifically, this invention describes optimized and standardized reagents and ultra-violet treatment to achieve an effective and highly reproducible nucleic acid inactivation prior to NAT without substantially affecting the performance of the assay. More specifically, this nucleic acid inactivation process resulted in a reduction of up to four logs of the background signal associated with the PCR (polymerase chain reaction) amplification of DNA contaminating PCR reagents. This optimized and standardized method is also adaptable for use with NAT technologies other than PCR.
La présente invention a trait à des réactifs exempts d'acides nucléiques contaminants détectables permettant la réalisation d'amplification et/ou de détection d'acides nucléiques à sensibilité et spécificité élevées. L'invention a trait à une amélioration dans la technologie de l'inactivation d'acides nucléiques préalablement au test d'acides nucléiques en vue d'éviter des résultats faux positifs. Plus particulièrement, la présente invention a trait à des réactifs optimisés et normalisés et un traitement par rayonnement ultraviolet pour obtenir une inactivation efficace et hautement reproductible de l'inactivation d'acides nucléiques préalablement au test d'acides nucléiques sans affecter sensiblement la performance du dosage. De manière plus spécifique, ce processus d'inactivation d'acides nucléiques a entraîné une réduction d'au moins quatre logs du signal de fond associé à l'amplification par la polymérase (PCR) de réactifs PCR contaminant d'ADN. Ce procédé optimisé et normalisé est également adaptable pour une utilisation avec des technologies de test d'acides nucléiques autres que la PCR.
