Abstract A method that is direct and stereospecific is provided for the production of 2'-deoxyadenosine derivatives and related analogs. The method comprises glycosylation of the sodium salt of 2,6-dichloropurine or 6-chloropurine and ammonolysis of the glycosylate to obtain the corresponding 2-chloro-2'-deoxyadenosine or 2'-deoxyadenosine. The method is useful for the production of the 2'deoxyadenosine compounds which in turn are useful cytotoxic agents.
