A method for the production of the N-terminal four kringle-containing fragment of hepatocyte growth factor (NK4) by expression of a nucleic acid encoding said NK4 in a microbial host cell, isolation of inclusion bodies containing said NK4 in denatured form, solubilization of the inclusion bodies and naturation of the denatured NK4, characterized in that solubilization and naturation are performed at pH 7-9 in phosphate buffered solution, provides NK4 in high purity and high yield.

L'invention porte sur une méthode d'obtention du fragment N-terminal du facteur de croissance à 4 kringle de l'hépatocyte (NK4) comportant les étapes suivantes: expression d'un acide nucléique codant pour ledit NK4 dans une cellule hôte microbienne; isolement des corps d'inclusion contenant ledit NK4 sous forme dénaturée; solubilisation des corps d'inclusion; et naturation du NK4 dénaturé, les étapes de solubilisation et de naturation s'effectuant sous un pH de 7-9 dans une solution à tampon phosphate. On obtient ainsi un NK4 de grande pureté avec un fort rendement.
