The invention relates to a method for the real-time quantification of a nucleic acid (10) by means of a nucleic acid duplicating reaction in the presence of at least one probe (18) that specifically hybridizes with said nucleic acid (10) or the complementary counter-strand (8) thereof. Hybridization is canceled by means of enzymatic degradation of the probe (18) if the nucleic acid that is to be detected is present during the nucleic acid duplicating reaction, canceling of the hybridization process being detected in an electrochemical manner. Said probe (18) comprises at least one electrochemically detectable marker (20) by means of which electrochemical detection occurs, said electrochemical detection being direct proof of an electrochemical reaction by discharging an electrical signal on an electrode. The intact probe (18) is prevented from accessing the electrode (30) while the degradation products of the probe (18) are not.
L'invention concerne un procédé d'analyse quantitative en temps réel d'un acide nucléique (10) par l'intermédiaire d'une réaction de multiplication de l'acide nucléique. Ce procédé fait intervenir au moins une sonde (18) formant spécifiquement une hybridation avec l'acide nucléique (10) ou avec son brin complémentaire (8). Cette hybridation est supprimée par dégradation enzymatique de la sonde (18) en présence de l'acide nucléique à détecter lors de la réaction de multiplication de l'acide nucléique, la suppression de l'hybridation étant détectée de façon électrochimique. La sonde (18) comprend au moins une substance de marquage (20) détectable de façon électrochimique au moyen de laquelle la détection électrochimique peut être effectuée. Cette détection électrochimique est une détection directe d'une réaction électrochimique par dérivation d'un signal électrique au niveau d'une électrode. L'accès de la sonde (18) intacte à l'électrode (30) est empêché, mais pas l'accès des produits de dégradation de la sonde (18).
