A method for the recombination of a gene is disclosed. The method involves the design of unpaired forward and reverse primers having homology to the 5' end of one template and to the 3 end of another template. Short primer extension periods results in a recombined template having paired 5' and 3' ends that can then be amplified. The amplified sample is devoid of any parental template.

La présente invention a trait à un procédé pour la recombinaison d'un gène. Le procédé consiste en la conception d'amorces non appariées avant et inverse présentant une homologie de l'extrémité 5' d'une matrice et à l'extrémité 3' d'une autre matrice. Des périodes courtes de l'extension des amorces permet d'obtenir une matrice recombinée présentant des extrémités 5' et 3' appariées qui peuvent ensuite être amplifiées. L'échantillon amplifié est exempt de toute matrice d'origine.
