A process for the manufacture of omeprazole or esomeprazole from pyrmethyl alcohol via pyrmethyl chloride and pyrmetazole characterized in that the whole reaction sequence is carried out without any isolation or purification of intermediates. Further, the reaction is carried out in a solvent system common for the whole reaction sequence and inert to the reactants formed during the process and used in the process and comprises a water immiscible organic solvent and a specified amount of water.

L'invention concerne un procédé de fabrication d'oméprazole ou d'ésoméprazole à partir d'alcool pyrméthylique par l'intermédiaire de chlorure pyrméthylique et de pyrmétazole. Le procédé est caractérisé en ce que la séquence de réaction entière est effectuée sans aucun isolement ni purification d'intermédiaires. De plus, la réaction est effectuée dans un système de solvants commun pour la séquence de réaction entière et inerte pour les réactifs formés au cours du procédé et utilisés dans le procédé, ledit système comprenant un solvant organique immiscible dans l'eau ainsi qu'une quantité spécifiée d'eau.
