A method of microbe culturing, in which a microbe capable of producing at least either a highly unsaturated fatty acid or a compound containing a highly unsaturated fatty acid as a constituent fatty acid is cultured in a culture medium containing at least a carbon source and a nitrogen source by means of an aerated spinner culturing apparatus capable of regulation and control of agitation power and aeration rate. In particular, there is provided a method of fungus culturing wherein while a mechanical agitation of <= 269 W/m3 agitation power per liquid quantity is performed for a given period of time from culturing initiation, at least either the maximum aeration rate or the maximum agitation power required is regulated and controlled after the passage of the given period of time within such a range as to satisfy the relationships of, when P refers to agitation power required (W), V to liquid quantity (m3) and Vs to aeration linear velocity (m/sec), KLA(=(P/V)0.95Vs0.67) >= 59, aeration linear velocity parameter Vs0.67 >= 0.075 and agitation power requirement parameter (P/V)0.95 >= 203.
