The presence or absence of an organism in a sample is detected by isolating the organism from the sample by a suitable affinity matrix, releasing the organism from the affinity matrix, rupturing the cells of the organism to release total nucleic acid and hydrolyzing or digesting the total nucleic acid to form mononucleotides or individual free nucleic acid bases and inorganic phosphate to form an analyte solution, and assaying the analyze solution for the at least one presence of free nucleic acid base or inorganic phosphate to thereby determine whether the organism was present in the sample.

On détecte la présence ou l'absence d'un organisme dans un échantillon en isolant l'organisme de l'échantillon au moyen d'un matrice d'affinité appropriée, en libérant l'organisme de ladite matrice par la rupture des cellules de l'organisme de manière que l'acide nucléique total soit libéré et en hydrolysant ou en digérant ledit acide nucléique pour former des mononucléotides ou des bases d'acide nucléique libres et du phosphate inorganique et produire une solution d'analyte, et en analysant ladite solution pour rechercher au moins la présence de la base d'acide nucléique libre ou de phosphate inorganique ou les deux, et déterminer si l'organisme est présent dans l'échantillon.
