A method of detecting a reactive metabolite which involves the reaction step wherein a metabolic reaction of a test compound is conducted in the presence of two labeled nucleophiles and the detection step wherein the label of the nucleophile bonded to the reactive metabolite is detected; and a method of detecting a reactive metabolite which further involves the separation step wherein a liquid layer separation is conducted by using ammonium acetate to give an organic layer after the completion of the reaction step as described above. Use of these detection methods makes it possible to quantitatively detect a reactive metabolite while using convenient preparation procedures. Moreover, it becomes possible thereby to avoid the formation of false positive signals and lessen the formation of false negative signals. As a result, a test compound can be more accurately and more efficiently screened.
La présente invention concerne un procédé de détection d~un métabolite réactif qui implique l'étape de réaction dans laquelle une réaction métabolique d'un composé d'essai est effectuée en présence de deux nucléophiles marqués et l'étape de détection dans laquelle le marqueur du nucléophile lié au métabolite réactif est détecté ; et un procédé de détection d~un métabolite réactif qui implique en outre l'étape de séparation dans laquelle une séparation d~une phase liquide est effectuée en utilisant de l'acétate d'ammonium pour donner une phase organique après avoir effectué l'étape de réaction telle que décrite ci-dessus. L~utilisation de ces procédés de détection rend possible la détection quantitative d~un métabolite réactif tout en utilisant des procédures de préparation pratiques. En outre, il devient de ce fait possible d'éviter la formation de signaux positifs faux et de diminuer la formation de signaux négatifs faux. En conséquence, un composé d'essai peut être analysé de manière plus précise et plus efficace.
