A method for inducing and enhancing protective and/or therapeutic immunity in a mammal to HSV includes the steps of at least one immunization with an effective amount of a DNA vaccine composition which comprises a first nucleic acid molecule comprising a DNA sequence encoding the HSV type 1 or type 2 gD protein, and a second nucleic acid molecule comprising a DNA sequence encoding each Interleukin-12 heterodimer subunit. The method also comprises at least one subsequent immunization with an effective amount of a protein vaccine composition which comprises the HSV type 1 or type 2 gD protein; and the IL-12 heterodimer. A local anesthetic may be included in the DNA vaccine composition in an amount that forms one or more complexes with the nucleic acid molecules. When provided to the mammal in suitable effective dosages according to this protocol, the vaccine compositions used in this method produce an unexpectedly good protective and/or therapeutic immune response against HSV in an immunized mammal.
L'invention se rapporte à une méthode visant à induire et à renforcer chez un mammifère l'immunité protectrice et/ou thérapeutique face au HSV. Ladite méthode consiste en au moins une immunisation avec une quantité efficace d'une composition vaccinale d'ADN qui contient une première molécule d'acide nucléique comportant une séquence d'ADN codant la protéine gD du HSV de type 1 ou de type 2, et une seconde molécule d'acide nucléique comportant une séquence d'ADN codant chaque sous-unité hétérodimère de l'Interleukine-12. Ladite méthode consiste également en au moins une immunisation subséquente avec une quantité efficace d'une composition vaccinale protéique qui contient la protéine gD du HSV de type 1 ou de type 2; et l'hétérodimère IL-12. Il est possible d'ajouter un anesthésique local à la composition vaccinale d'ADN en quantité permettant la formation d'un ou de plusieurs complexes avec les molécules d'acides nucléiques. Lorsqu'elles sont administrées à un mammifère à des dosages efficaces appropriés conformément au protocole décrit, les compositions vaccinales utilisées par ladite méthode génèrent une réponse immunitaire anti-HSV, protectrice et/ou thérapeutique, étonnamment bonne, chez le mammifère immunisé.
