2066632 9105042 PCTABS00004 A process for isolating, cloning and maintaining large genomic fragments of chromosomal DNA. This process includes the steps of introducing a marker into the chromosomal DNA to be cloned, inserting double-strand breaks into the chromosome followed immediately by combining the cells containing the chromosome with the fragmented marker to another cell line capable of harboring extrachromosomal DNA elements, and subsequently isolating genomic fragments by separating the extrachromosomal DNA from the chromosomal DNA of the other cell line.
