ABSTRACT Disclosed is a method by which expression of any gene in an intact, unfused form can be achieved. The gene to be expressed is amplified using Polymerase Chain Reaction (PCR) starting from the second codon of the particular gene and the resulting product is ligated to the ATG codon which is provided by the Ncol site of the expression vector. This results in the expression of the gene in an intact, unfused form with no extra amino acids. Using this strategy high level expression of Plasmodium falciparum hypoxanthine guanine phosphoribosyl transferase (Pf HGPRT) gene, in E.coli, has been achieved.
