A method of identifying antigens which are capable of being endogenously processed by a cellular secretory pathway comprising: introducing an antigen into a donor cell lacking in MHC class I molecules, incubating in an in vitro medium the donor cells, primed cytotoxic T lymphocytes having specificity for the antigen, and target cells which express MHC class I molecules and are labelled with a detectable intracellular marker, under suitable conditions such that the donor cells remain intact, and measuring the amount of detectable marker released into the incubation medium. A method of assaying a medium for the presence of a substance that affects processing of an endogenously processed antigen in a cellular secretory pathway; and a method of characterizing a tumor or viral antigen capable of being endogenously processed by a cellular secretory pathway.
Procédé d'identification d'antigènes pouvant être traités de manière endogène par une voie sécrétoire cellulaire, consistant à: introduire un antigène dans une cellule donneuse dépourvue de molécules du complexe majeur d'histocompatibilité (CMH) de classe I, incuber, dans un milieu in vitro les cellules donneuses, des lymphocytes T cytotoxiques amorcés spécifiques de l'antigène, ainsi que des cellules cibles qui expriment les molécules CMH de classe I et qui sont marquées au moyen d'un marqueur intracellulaire détectable, dans des conditions telles que les cellules donneuses restent intactes, et mesurer la quantité de marqueur détectable libéré dans le milieu d'incubation. Un procédé de dosage d'un milieu pour détecter la présence d'une substance affectant le traitement d'un antigène traité de manière endogène dans une voie sécrétoire cellulaire est également décrit, ainsi qu'un procédé de caractérisation d'un antigène tumoral ou viral pouvant être traité de manière endogène par une voie sécrétoire cellulaire.
