A method of increasing the affinity and/or decreasing the off-rate of a given TCR specific for a given target pMHC, comprising creating a plurality of TCRs having an .alpha. chain CDR2 sequence and/or a .beta. chain CDR2 sequence different from the corresponding CDR2 sequence(s) of the given TCR but having the same .alpha. and .beta. CDR1 and CDR3 sequences as the given TCR, determining the affinity and/or off-rate of members of said plurality of TCRs for the target pMHC, and selecting one or more members having at least a 10- fold greater affinity for the target pMHC than the given TCR and/or a 10-fold slower off-rate for the target pMHC than the given TCR.

L'invention concerne un procédé permettant d'accroître l'affinité et/ou de diminuer la dissociation d'un TRC donné spécifique pour un pMHC cible donné et consistant à créer une pluralité de TCR possédant une séquence CDR2 à chaîne .alpha. et/ou une séquence CDR2 à chaîne .beta. différentes des séquences CDR2 correspondantes du TCR donné mais possédant les mêmes séquences CDR1 et CDR3 à chaînes .alpha. et .beta. que le TCR donné, à déterminer l'affinité et/ou la dissociation d'éléments de la pluralité de TCR destinés au pMHC cible et à sélectionner un ou plusieurs éléments possédant au moins une affinité 10 fois supérieure pour le pMHC cible que le TCR donné et/ou une dissociation plus lente pour le pMHC donné que le TCR donné.
