The present invention relates to a method of interfering with gene expression in a mammalian past-implantation embryo comprising the step of introducing siRNA molecules or shRNA molecules or DNA molecules encoding siRNA or shRNA molecules wherein said siRNA molecules or said shRNA molecules have a region of identity or of substantial identity of at least 15 nucleotides with the gene(s) the expression of which is to be interfered with into cells of said post- implantation embryo. Additionally, the present invention pertains to a method of performing a genomic-wide functional screen comprising introducing variety of siRNA molecules or shRNA molecules or DNA molecules encoding a variety of siRNA or shRNA molecules wherein said different siRNA or shRNA molecules have a region of identity or substantial identity of at least 15 by with different genes into a mammalian post-implantation embryo.
