The present invention provides a method for measurement of concentration of fluorophores in turbid media. The method uses a fluorescence plus reflectance technique to measure quantitatively the iin vivo concentration of a fluorescent drug independent of the tissue optical properties. The method involves using the reflectance measurement as a means of correcting for tissue scattering and absorption in the fluorescence measurement. Light directed to the tissue by means of an optical fiber to excite the fluorophores. Fluorescence from the fluorophores is measured by another fiber at a distance (D1) from the source in the turbid medium, while the excitation light is simultaneously collected at a distance (D2) from the source. The ratio fluorescence/reflectance will be approximately constant for a fixed concentration of the fluorophore and a judicious choice of D1 and D2, regardless of the optical properties of the tissue. The ratio of fluorescence to reflectance on a tissue sample is compared to a calibration curve for a range of fluorophore concentrations, in order to estimate the in vivo concentration in the turbid medium.

La présente invention concerne un procédé de mesure de la concentration de fluorophores dans des milieux troubles. On utilise dans ce procédé une technique de fluorescence et de réflectance pour mesurer quantitativement la concentration in vivo d'un médicament fluorescent indépendamment des propriétés optiques du tissu. Le procédé implique l'utilisation de la mesure de réflectance comme moyen pour corriger la diffusion et l'absorption par le tissu qui apparaissent dans la mesure de fluorescence. La lumière dirigée sur le tissu par une fibre optique vise à exciter les fluorophores. La fluorescence émise par les fluorophores est mesurée par une autre fibre à une distance (D1) de la source dans le milieu trouble, la lumière d'excitation étant simultanément collectée à une distance (D2) de la source. Le rapport fluorescence/réflectance est approximativement constant pour une concentration fixe du fluorophore et un choix judicieux des distances D1 et D2, quelles que soient les propriétés optiques du tissu. Le rapport entre la fluorescence et la réflectance sur un échantillon de tissu est comparé à une courbe d'étalonnage associée à une fourchette de concentrations de fluorophore, afin d'estimer la concentration in vivo dans le milieu trouble.
