A method of detecting or measuring homocysteine in a sample which comprises: (a) the step of treating a D-amino acid in the sample with a D-amino acid converting enzyme to thereby convert the D-amino acid into a substance not acting as the substrate of D-amino acid oxidase or D-amino acid acetyltransferase; (b) the step of reducing homocysteine in the sample with a thiol compound; (c) the step of treating the thus reduced homocysteine with methyltransferase and a methyl donor to thereby newly form a D-amino acid; and (d) treating the thus formed D-amino acid with the D-amino acid oxidase or the D-amino acid acetyltransferase in the presence of an SH reagent to thereby derive it into hydrogen peroxide and then coloring the thus formed hydrogen peroxide with the use of an oxidative color developing agent.
L'invention concerne un procédé de détection ou de mesure d'homocystéine dans un échantillon, qui comprend les étapes consistant: (a) à traiter un acide aminé de type D dans l'échantillon au moyen d'une enzyme de conversion d'acide aminé de type D afin de convertir ledit acide aminé de type D en une substance ne faisant pas office de substrat d'oxydase d'acide aminé de type D ou d'acétyltransférase d'acide aminé de type D; (b) à réduire l'homocystéine dans l'échantillon au moyen d'un composé thiol; (c) à traiter l'homocystéine ainsi réduite au moyen d'une méthyltransférase et d'un donneur de méthyle afin que soit de nouveau formé un acide aminé de type D; et (d) à traiter l'acide aminé de type D ainsi formé au moyen de l'oxydase d'acide aminé de type D ou de l'acétyltransférase d'acide aminé de type D en présence d'un réactif SH afin de le dériver en peroxyde d'hydrogène et à colorer ledit peroxyde d'hydrogène ainsi formé au moyen d'un agent oxydant de développement de couleur.
