The present inventors provide methods for modifying the isoelectric point of an antibody while retaining its antigen-binding activity, comprising modifying the charge of at least one exposable amino acid residue on the surface of the complementarity determining region (CDR). The present invention also provides methods for purifying multispecific antibodies, comprising modifying isoelectric point, and methods for improving the plasma pharmacokinetics of antibodies with a modified isoelectric point. The present invention further provides antibodies with a modified isoelectric point, pharmaceutical compositions comprising the antibodies as an active ingredient, and methods for producing the antibodies and compositions.
Cette invention concerne un procédé permettant à la fois de modifier le point isoélectrique d'un anticorps et de conserver l'activité de liaison à l'antigène de ce dernier. Ce procédé consiste à modifier la charge d'au moins un reste d'acide aminé pouvant être exposé sur la surface de la région déterminant la complémentarité (CDR). Cette invention concerne également un procédé permettant de purifier un anticorps multispécifique par modification de son point isoélectrique, ainsi qu'un procédé consistant à améliorer la cinétique des médicaments dans le plasma d'un anticorps présentant un point isoélectrique modifié. L'invention concerne également un anticorps présentant un point isoélectrique modifié, une composition médicamenteuse comprenant l'anticorps en tant qu'ingrédient actif ainsi qu'un procédé de production correspondant.
