The invention relates to a method of preparing 2-amino-6-alkyl-amino-4,5,6,7- tetrahydrobenzothiazoles having formula (I), wherein the asterisk (*) represents an asymmetrical carbon and R1 is alkyl C1-C6, the enantiomers or mixtures thereof, the pharmaceutically-acceptable solvates, hydrates or salts of same. The inventive method comprises the following steps consisting in: (a) reacting a compound (II) with a secondary amine, optionally in the presence of an acid and a solvent 1, in order to form an enamine; (b) optionally removing the acid and solvent 1 and, subsequently, reacting the enamine with sulphur in the presence of a solvent 2; and (c) reacting the previously obtained compound with cyanamide in order to obtain compound (I). Compounds having formula (I) include pramipexol which is a compound with dopamine D2 agonist activity that can be used for the treatment of Parkinson's disease and schizophrenia.
L'invention concerne un procédé de préparation d'un composé 2-amino-6-alkyl-amino-4,5,6,7-tétrahydrobenzotiazoles de formule (I) dans laquelle l'astérisque (*) représente carbone asymétrique et R1 représente alkyle C1-C6 ; ses énantiomères ou des mélanges de ceux-ci, ses solvates, hydrates ou sels acceptables sur le plan pharmaceutique. Ce procédé consiste : (a) à faire réagir un composé de formule (II) avec une amine secondaire, éventuellement en présence d'un acide et d'un dissolvant 1, pour former une énamine ; (b) éventuellement à retirer ledit acide et ledit dissolvant 1, puis à faire réagir ladite énamine avec du soufre en présence d'un dissolvant 2 ; et (c), à faire réagir le composé précédemment obtenu avec du cyanamide afin d'obtenir le composé de formule (I). Parmi les composés de formule (I), on trouve le pramipexol, un composé présentant une activité agoniste D-2 de la dopamine, utile pour le traitement de la maladie de Parkinson et la schizophrénie.
