Antibodies are recovered and isolated from the sera of a number "n" of patients each of which have or have had the same given disease so that each of the "n" patients has, within a large number of different antibodies, some antibodies uniquely associated with the same disease of interest. The antibodies of a first patient are bound to a support surface. To carry out initial screening libraries of molecules, such as peptides displayed on the surface of bacteriophage, are brought into contact with the bound antibodies of the first patient under conditions where binding will occur. Unbound molecules (peptides) are washed away and bound molecules (peptides) on the bacteriophage are identified. Secondary screening is then carried out by extracting and labeling the antibodies of a second patient and using the labeled antibodies to probe the molecules (peptides) isolated in the initial screening.
On récupère et on isole les anticorps extraits de sérums de "n" patients qui ont, ou qui ont eu, la même maladie donnée, de sorte que chacun des "n" patients a, à l'intérieur d'une large gamme d'anticorps différents, certains anticorps uniquement associés à la maladie en question. Les anticorps d'un premier patient sont fixés sur une surface de support. Pour effectuer un examen initial, on amène des bibliothèques de molécules, telles que les peptides présents sur la surface d'un bactériophage en contact avec les anticorps liés du premier patient dans des conditions où une fixation va avoir lieu. Les molécules non liées (peptides) sont éliminées par lavage et les molécules liées (peptides) sur le bactériophage sont identifiées. On effectue ensuite un examen secondaire en extrayant et en marquant les anticorps du second patient et en utilisant les anticorps marqués pour sonder les molécules (peptides) isolées au cours de l'examen initial.
