A method of preventing, reducing or reversing nephrotoxicity or renal dysfunction induced by administration of a cyclosporin or tacrolimus to a mammalian patient. The method comprises the co-administration to the patient, either before, together with or after cyclosporin or tacrolimus administration, of a pharmaceutical composition containing an effective amount of pentosan polysulfate (PPS) or a pharmaceutically acceptable salt thereof. The oral route of administration is preferred. The total daily dosage of PPS or PPS salt ranges from about 2 to about 50 mg/kg of patient body weight, or about 140 to about 3,500 mg per day in adult human patients. Also disclosed are a method of providing immunosuppressive therapy to a patient while avoiding cyclosporin or tacrolimus-induced nephrotoxicity, and combination pharmaceutical compositions to be used in such therapy.
L'invention concerne un procédé qui permet de prévenir, de réduire ou d'éliminer la néphrotoxicité ou le dysfonctionnement rénal résultant de l'administration de cyclosporine ou de tacrolimus à un patient mammifère. Le procédé consiste à co-administrer au patient, avant ou avec l'administration de cyclosporine ou de tacrolimus, une composition pharmaceutique renfermant une quantité efficace de pentose polysulfate ou un sel pharmaceutiquement acceptable de ce produit. De préférence, l'administration est par voie orale. La dose quotidienne du produit considéré ou du sel pharmaceutiquement acceptable de ce produit oscille entre environ 2 et environ 50 mg/kg de poids corporel du patient, ou entre environ 140 et environ 3 500 mg/jour chez les patients adultes. L'invention concerne également un procédé qui permet d'assurer au patient un traitement aux agents immunosuppresseurs tout en évitant la néphrotoxicité résultant de l'administration de cyclosporine ou de tacrolimus, et elle concerne aussi des compositions pharmaceutiques destinées à être utilisées en combinaison avec ledit traitement.
