Provided herein are methods and compositions for producing Factor VIII proteins. Such methods include introducing into a cell a nucleic acid molecule encoding a Factor VIII protein operably linked to a promoter, wherein the promoter is characterized by the ability to produce commercially viable Factor VIII protein; and incubating the cell under conditions for producing commercially viable Factor VIII protein. Also provided are nucleic acid molecules which encode a Factor VIII protein operably linked to a Chinese hamster elongation factor 1-.alpha. (CHEF1) promoter, which may be used in the methods provided herein.
La présente invention concerne des procédés et des compositions de production de protéines de type Facteur VIII. Lesdits procédés consistent à introduire dans une cellule une molécule d'acide nucléique codant pour une protéine de type Facteur VIII qui présente une liaison fonctionnelle avec un promoteur, le promoteur étant caractérisé par la capacité de produire une protéine de type Facteur VIII commercialement viable, puis à incuber la cellule dans des conditions permettant de produire une protéine de type Facteur VIII commercialement viable. La présente invention concerne également des molécules d'acide nucléique qui codent pour une protéine de type Facteur VIII présentant une liaison fonctionnelle avec le promoteur du facteur d'élongation 1-.alpha. du hamster chinois (CHEF1), lesdites molécules étant susceptibles d'être utilisées dans les procédés décrits dans ce document.
