A method for producing microbeads comprising an active component encapsulated within a gelled polymer matrix comprises the steps of providing a suspension of denatured whey protein and an active component, treating the suspension to gen-erate microbeads, and immediately curing the microbeads by acidification. The microbeads are discrete droplets of gelled whey protein having an average diameter in the micron range (for example, from 80 to 500 microns) and which, suitably have a general-ly spherical shape. The microbeads are capable of surviving passage through the stomach, and delivering the encapsulated active agent in the instestine. Ex-vivo and in-vivo data shows that active agent encapsulated within microbeads retains its functionality upon delivery to the intestine, and that coating of the microcapsules allows targeted delivery of the active agent to the distal part of the intestine.

Cette invention concerne un procédé de production de microbilles comprenant un composant actif encapsulé à l'intérieur d'une matrice polymère gélifiée, ledit procédé consistant à fournir une suspension de protéine du petit-lait dénaturée et un composant actif, à traiter la suspension pour générer des microbilles, et à immédiatement polymériser les microbilles par acidification. Les microbilles sont des gouttelettes discrètes de protéine du petit-lait gélifiée ayant un diamètre moyen dans la gamme du micron (par exemple de 80 à 500 microns) et une forme généralement sphérique. Les microbilles sont capables de survivre au passage dans l'estomac, et de libérer l'agent actif encapsulé dans l'intestin. Les données obtenues ex-vivo et in-vivo montrent que l'agent actif encapsulé à l'intérieur des microbilles conserve sa fonctionnalité après libération dans intestin, et que l'enrobage des microcapsules permet l'administration ciblée de l'agent actif à la partie distale de l'intestin.
