It is intended to provide a technique whereby a target protein can be efficiently obtained as a normal type protein even in the case of a protein which can be hardly expressed by usual recombinant DNA techniques. A fused protein comprising a molecular chaperone, an intercalated protein having an activity of cleaving a peptide bond and a target protein is prepared. Then the target protein is cut out from the fused protein with the use of the function of the intercalated protein having an activity of cleaving a peptide bond. As examples of the intercalated protein having an activity of cleaving a peptide bond, an intein and its partial sequence protein can be cited.
La présente invention a trait à une technique permettant l'obtention efficace d'une protéine cible sous la forme d'une protéine de type normal même dans le cas d'une protéine qui peut difficilement être exprimée par les techniques d'ADN recombinant. La technique consiste en la préparation d'une protéine fusionnée comportant un chaperon moléculaire, une protéine intercalée présentant une activité de clivage d'une liaison peptidique et une protéine cible. Ensuite la protéine cible est découpée de la protéine fusionnée au moyen de la fonction de la protéine intercalée présentant une activité de clivage d'une liaison peptidique. On peut citer comme exemples de protéine intercalée présentant une activité de clivage d'une liaison peptidique, une intéine et sa protéine à séquence partielle.
