ABSTRACT The invention relates to a method of prophylaxis or treatment of atheromatosis in susceptible vertebrates comprising administration of managese-containing pharma- ceutical preparations in appropriate ratios of tryptophan to manganese to prevent development and progression of atheromatosis: both the manganese and tryptophan being given in cumulative amounts appropriate to the amount, frequency and ratios that it reflects in the changing degrees of susceptibility and imbalance in the metabolism of the affected individual as it returns to a normal metabolic state of the arterial intima and walls. The same therapeutic approach presents itself for use in individuals with premature atheromatosis of Hutchison- Gilford syndrome
