The invention provides a method for quantifying the cell-binding properties of a medical device. In practicing the method, a medical device having at least one type of binding agent is incubated with cells expressing a ligand having an affinity for the binding agent on the medical device. The cells that bind to the medical device are labeled with at least one marker. The marker is detected, and the quantity of cells bound to the medical device is determined. Alternatively, labeled cells having at least one type of ligand expressing an affinity for the at least one type of binding agent may be provided that are incubated with the medical device. The relative expression of the ligand on the cell line having an affinity for the binding agent is also determined. The method can be used to assess, in vitro, how well cells will bind to the medical device, thereby providing useful insight as to the effectiveness of the medical device to promote cell adherence to the device, prior to in vivo implantation of such devices.
L'invention concerne un procédé permettant de quantifier les propriétés de fixation cellulaires d'un dispositif médical. Ce procédé consiste à incuber un dispositif médical comportant au moins un type d'agent de fixation avec des cellules exprimant un ligand qui présente une affinité envers l'agent de fixation du dispositif médical, à marquer les cellules qui se fixent sur le dispositif médical avec au moins un marqueur, et à détecter le marqueur et la quantité de cellules fixées sur le dispositif médical. Dans un mode de mise en oeuvre différent, ce procédé consiste à prendre des cellules marquées présentant au moins un type de ligand exprimant une affinité envers au moins un type d'agent de fixation, et à les incuber avec le dispositif médical. L'expression relative du ligand par rapport à la lignée cellulaire présentant une affinité pour l'agent de fixation est également mesurée. Ce procédé peut servir à l'évaluation in vitro de la fixation des cellules sur le dispositif médical, offrant ainsi un aperçu utile de l'aptitude du dispositif médical à promouvoir l'adhérence cellulaire au dispositif, avant l'implantation in vitro de dispositifs de ce type.
