It is intended to provide a method of screening a candidate for a drug which is metabolized mainly by drug metabolizing enzymes CYP2D6, CYP2C9 and CYP2C19 with the use of a chimeric mouse carrying human hepatocytes transplanted thereinto, whereby it is judged whether or not a candidate for the drug is metabolized in a subject deficient in CYP2D6, CYP2C9 or CYP2C19. It is also intended to provide a method of judging whether or not a candidate for a drug induces or inhibits the activity of any one of drug metabolizing enzymes CYP1A2, CYP2C9, CYP2C19, CYP2D6 and CYP3A4 with the use of a chimeric mouse carrying human hepatocytes transplanted thereinto.
La présente invention concerne un procédé de criblage d~un candidat destiné à un médicament, principalement métabolisé par des enzymes métabolisant le médicament, CYP2D6, CYP2C9 et CYP2C19 et reposant sur l~utilisation d~une souris chimère porteuse d~hépatocytes humains transplantés, moyennant quoi on juge si un candidat destiné à un médicament est ou non métabolisé chez un sujet présentant une carence en CYP2D6, CYP2C9 ou CYP2C19. La présente invention concerne également un procédé permettant d'évaluer si un candidat destiné à un médicament induit ou inhibe l~activité de l~une quelconque des enzymes métabolisant le médicament CYP1A2, CYP2C9, CYP2C19, CYP2D6 et CYP3A4 en utilisant une souris chimère porteuse d~hépatocytes humains transplantés.
