To provide a method of efficiently separating protective components of Bordetella pertussis. On the basis of differences in adsorbability to calcium phosphate gel formed by adding calcium ions to a Bordetella pertussis culture in the presence of excess phosphate ions, protective components of Bordetella pertussis are separated from the Bordetella pertussis culture. Traditionally, protective components of Bordetella pertussis have been separated using different purification methods for the respective components. According to the present invention, the use of the same means of purification for all subject components makes it possible to purify each component with high efficiency and high recovery rate, an aspect very advantageous for industrial production. It is also possible to efficiently produce an improved purified pertussis component vaccine comprising an effective combination of pertussis filamentous hemagglutinin (FHA), pertactin (PRN, 69K- OMP), pertussis fimbriae (FIM) and pertussis toxin (PT).
Un procédé permet de séparer des éléments protecteurs de Bordetella pertussis. En fonction des différences de capacité d'absorption d'un gel de phosphate de calcium formé par addition d'ions calcium à une culture de Bordetella pertussis en présence d'ions phosphate excédentaires, on peut séparer des éléments protecteurs de Bordetella pertussis à partir d'une culture de Bordetella pertussis. On les séparait précédemment par un procédé de purification différent pour chacun d'entre eux et l'invention permet d'utiliser un même procédé de purification pour tous ces éléments et de recueillir chacun d'entre eux avec un rendement élevé, ce qui se révèle très avantageux en vue de leur production industrielle. On peut aussi produire de façon rentable un vaccin amélioré, à base d'éléments de pertussis modifiés, qui comprend une combinaison efficace d'hémagglutinine filamenteuse de pertussis (FHA), de pertactine (PRN, 69K-OMP), de pili bactériens de pertussis (FIM), et de toxine de pertussis (PT).
