The present invention is a method of separating nucleic acid molecules, in particular plasmid DNA, from contaminants, such as proteins, in a solution, which method comprises the following steps a) providing an aqueous adsorption solution, which comprises nucleic acids and a salt that forms lyotropic ions when dissolved; b) passing said solution over a matrix to adsorb the desired nucleic acids onto the matrix, which comprises an aromatic ring moiety and at least one thioether moiety; c) optionally washing the matrix; d) passing an aqueous eluent over said matrix to desorb the nucleic acid molecules therefrom, which eluent in addition to a salt that forms lyotropic ions also comprises a gradient of increasing ionic strength originating from an increasing concentration of salt that forms less lyotropic ions when dissolved than the ones present during adsorption; and e) isolating the fraction comprising the desired nucleic acid molecules.

L'invention porte sur un procédé de séparation de molécules d'acide nucléique, notamment d'ADN plasmidique, de contaminants, tels que des protéines, dans une solution. Ce procédé comprend plusieurs étapes consistant : (a) à fournir une solution d'adsorption aqueuse contenant des acides nucléiques et un sel qui forme des ions lors de leur dissolution, (b) à faire passer ladite solution sur une matrice afin d'adsorber les acides nucléiques désirés dans la matrice comprenant une fraction de noyau aromatique et au moins une fraction de thioéther, (c) facultativement, à laver la matrice, (d) à faire passer un éluant sur la matrice afin de désorber les molécules d'acide nucléique, cet éluant, en plus d'un sel formant des ions lyotropiques, comprenant également un gradient de force ionique accrue issue d'une concentration accrue de sel formant moins d'ions lyotropiques lors de leur dissolution que ceux présents au cours de l'adsorption, et (e) à isoler la fraction contenant les molécules d'acide nucléiques désirées.
