Methods of thrombolysis that allow the use of a fibrinolytic composition comprising reversibly inactivated acidified plasmin and the localized delivery of the plasmin to a vascular thrombotic occlusion are disclosed. Further disclosed is a method for administering a therapeutic dose of a fibrinolytic composition substantially free of plasminogen activator to a human or animal having a vascular thrombotic occlusion. The fibrinolytic composition includes a reversibly inactivated acidified plasmin substantially free of plasminogen activator. Intravascular catheter delivery of the fibrinolytic composition directly into or in the immediate vicinity of the thrombus is disclosed to minimize the systemic degradation of fibrin while retaining the maximum plasmin activity against the thrombus.
La présente invention concerne des procédés de thrombolyse qui font intervenir l'utilisation d'une composition fibrinolytique comprenant de la plasmine acidifiée inactivée de manière réversible, et l'administration locale de plasmine à une occlusion vasculaire thrombotique. Cette invention concerne également un procédé permettant l'administration à un être humain ou à un animal souffrant d'une occlusion vasculaire thrombotique, d'une dose thérapeutique d'une composition fibrinolytique pratiquement dépourvue d'activateur de plasminogène. La composition fibrinolytique comprend de la plasmine acidifiée inactivée de manière réversible pratiquement dépourvue d'activateur de plasminogène. L'invention concerne également l'administration par sonde intravasculaire de la composition fibrinolytique directement dans ou à proximité du thrombus, permettant de minimiser la dégradation systémique de la fibrine tout en maintenant une activité maximale de la plasmine contre le thrombus.
