A method of pharmaceutical therapy comprising the co-administration of any form of interferon or any derivative thereof with a low dose of ribavirin (less than 400 mg /day or less than 6 mg/kg/day), or related compound, where the ribavirin or related compound provides a clinically effective blood level in the portal circulation but a less than clinically effective blood level in the peripheral circulation, to thereby provide a systemic effect of interferon throughout the body but a selective effect of ribavirin in the liver. The method also provides for the co-administration of any form of interferon or any derivative thereof with a high dose of ribavirin (preferably from 400-800 mg/day), or related compound, where the ribavirin or related compound is administered as a slow-release formulation such that it also provides a sustained virologic response in a patient and reduced side effects. The method also provides for the co-administration of an antioxidant or other membrane protective agent with both the interferon and ribavirin such that the hepatoprotective activity of the antioxidant or other membrane protective agent complements the virucidal effect of the interferon and ribavirin. The antioxidant or other membrane protective agent may be administered as a systemic or a low-dose, slow-release, liver-selective formulation.
L'invention concerne une méthode de thérapie pharmaceutique consistant à co-administrer une forme quelconque d'interféron ou d'un dérivé quelconque de celui-ci avec une faible dose de ribavirine (moins de 400 mg/jour ou moins de 6 mg/kg/jour), ou d'un composé correspondant. La ribavirine ou le composé correspondant permet d'obtenir un niveau sanguin cliniquement efficace dans la circulation portale mais cliniquement moins efficace dans la circulation périphérique afin de produire un effet systémique de l'interféron d'un bout à l'autre du corps et un effet sélectif de la ribavirine dans le foie. Ladite méthode consiste à co-administrer une forme quelconque d'interféron ou d'un dérivé quelconque de celui-ci avec une dose élevée de ribavirine (de préférence, à partir de 400-800 mg/jour) ou d'un composé correspondant. La ribavirine ou le composé correspondant est administré sous forme de formulation à libération lente de sorte qu'elle ou il fournit une réponse virologique prolongée chez un patient et réduit les effets secondaires. Cette méthode consiste également à co-administrer un antioxydant ou un autre agent membranaire protecteur avec à la fois l'interféron et la ribavirine de sorte que l'activité hépatoprotectrice de l'antioxydant ou de l'autre agent membranaire protecteur complète l'effet virucide de l'interféron et de la ribavirine. L'antioxydant ou l'autre agent membranaire protecteur peut être administré sous forme de formulation à libération prolongée, systémique ou à faible dos et sélective dans le foie.
