Methods for using Zot or zonulin as an antigen specific inhibitor of APC activity and lymphocyte proliferation, being primarily useful in the field of immunoregulation and immunotherapy are described. Specifically, Zot and zonulin inhibit antigen presenting cell-mediated antigen-specific lymphocyte proliferation in a dose dependent manner. This effect is associated with the presence of a macrophage surface receptor to which Zot binds in a specific and saturable way. This down-regulation of the immune response is, at least in part, associated with a decreased uptake of antigen.

Cette invention a trait à des techniques d'utilisation de la toxine de zonula occludens (Zot) ou de la zonuline comme inhibiteur, spécifique d'antigène, de l'activité de cellules porteuses d'antigènes (APC) et de la prolifération lymphocytaire, ces substances s'utilisant principalement dans les domaines de la régulation immunitaire et de l'immunothérapie. Concrètement, La toxine Zot et la zonuline inhibent la prolifération lymphocytaire à spécificité antigénique induite par une cellule porteuse d'antigènes et ce, proportionnellement à la dose administrée. Cet effet est associé à la présence d'un récepteur de surface de macrophage auquel se fixe la toxine Zot de manière spécifique et saturable.
